Mass Spectrometric Analysis of Glycosphingolipid Antigens
Glycosphingolipids (GSL's) belong to the glycoconjugate class of biomacromolecules, which bear structural information for significant biological processes such as embryonic development, signal transduction, and immune receptor recognition(1-2). They contain complex sugar moieties in the form of isomers, and lipid moieties with variations including fatty acyl chain length, unsaturation, and hydr...
متن کاملMass-spectrometric analysis of glycerophospholipid metabolism
This thesis consists of five parts. In the first part, an automated method for quantitative analysis of phospholipid compositions of cells and tissues by liquid chromatographymass spectrometry was developed. In the second part, this method was applied to investigate brain lipid compositions of patients with progressive epilepsy with mental retardation (EPMR), caused by mutations in the CLN8 gen...
متن کاملMass spectrometric analysis of glucose-modified ribonuclease.
RNase A (1 mM) was incubated with glucose (0.4 M) at 37 degrees C for up to 14 days in phosphate buffer (0.2 M, pH 7.4), digested with trypsin and analysed by LC-MS. The major sites of fructoselysine formation were Lys(1), Lys(7), Lys(37) and Lys(41). Three of these sites (Lys(7), Lys(37) and Lys(41)) were also the major sites of N epsilon-(carboxymethyl)lysine formation.
متن کاملMass spectrometric analysis of protein–ligand interactions
The interactions of small molecules with proteins (protein-ligand interactions) mediate various biological phenomena including signal transduction and protein transcription and translation. Synthetic compounds such as drugs can also bind to target proteins, leading to the inhibition of protein-ligand interactions. These interactions typically accompany association-dissociation equilibrium accor...
متن کاملGoing beyond electrospray: mass spectrometric studies of chemical reactions in and on liquids
There has been a burst in the number and variety of available ionization techniques to use mass spectrometry to monitor chemical reactions in and on liquids. Chemists have gained the capability to access chemistry at unprecedented timescales, and monitor reactions and detect intermediates under almost any set of conditions. Herein, recently developed ionization techniques that facilitate mechan...
